Paprastosios saulėgrąžos sėklų apdorojimo elektromagnetinėmis bangomis ir šalta plazma poveikis sėklų daigumui, daigų morfologijai ir proteomai
Sėklų apdorojimas šalta plazma (ŠP) ar elektromagnetiniais laukais (EML) yra moderni žemėws ūkio technologija, taikoma siekiant stimuliuoti augalų daigumą ir pagerinti savybes. Atlikta daug tyrimų, liudijančių tokių poveikių efektyvumą didinant daigumą ir užauginamo derliaus kiekį, tačiau augalų atsako poveikiui molekuliniai mechanizmai nėra žinomi. Norint išsiaiškinti atsako stresoriams molekulinius mechanizmus, galinčius padėti suprasti, kaip augalas atsako į apdorojimą, įvertinti stresorių sukelti skirtingos baltymų raiškos lygiai modelinio augalo – paprastosios saulėgrąžos (Helianthus annuus) ʽNyķrségi feketeʼ kultivare. Ištirta sėklų apdorojimo prieš sėją vakuumu (7 min.), radijo dažnio EML (5-15 min.) ir ŠP (2-7 min.) poveikis daigumui ir daigų vystymuisi. Daigumo testai atlikti in vitro ir kasetėse. Gauti rezultatai, kad sėklų apdorojimas ŠP ir EML prieš sėją nepakeitė galutinės sudygusių sėklų išeigos, tačiau apdorojimas vakuumu, EML10 ir EML15 didino dygimo greitį, o apdorojimas vakuumu, ŠP5, ŠP7, EML5 ir EML10 mažino sėklų populiacijos dygimo vienodumą. Apdorojimas turėjo poveikį daigų, išaugusių iš ŠP7 ir EML15 apdorotų sėklų, morfologijai, kuris pasireiškė kaip stiebų aukščio ir svorio sumažėjimas bei lapų svorio padidėjimas. Pakitusi baltymų raiška saulėgrąžų daigų lapuose įvertinta naudojant 2D gelio elektroforezę. Iš 104 skirtinga raiška pasižymėjusių proteoformų, buvo nustatyta 49 ir 38 proteoformų padidėjusi raiška bei 26 ir 33 proteoformų sumažėjusi raiška, atitinkamai ŠP ir EML paveiktoms grupėms. Atlikus analizę, išryškėjo 3 pagrindinės baltymų raiškos hierarchiniame klasteryje grupės. Masių spektrometrijos būdu nustatyti baltymai, dalyvaujantys pirminiame ar baltymų metabolizme bei atsako į stresą reakcijose. Tarp 40 identifikuotų proteoformų dominavo chloroplastų baltymai, atliekantys svarbias fotosintezei funkcijas.
Seed treatment with cold plasma (CP) or electromagnetic field (EMF) is a modern agricultural technology for stimulation of plant germination and performance. Numerous studies demonstrated the effectiveness of such treatments for enhancing germination and growth, however the molecular basis of plants response to treatments remains elusive. In order to gain an insight into the molecular mechanisms underlying effect of stressors on plants, we estimated changes induced in differential protein expression of the model plant – sunflower (Helianthus annuus) ʽNyķrségi feketeʽ cultivar. We studied the effects of pre-sowing seed treatment, using vacuum (7 min), radio-frequency EMF (5-15 min) and CP (2-7 min), on germination and growth. The germination tests were performed both in vitro and in substrate. The obtained results indicated that the treatments with CP and EMF had no effect on the proportion of germinated seeds but vacuum and EMF (10 and 15 min) treatments increased germination rate, while vacuum, CP (5 and 7 min) and EMF (5 and 10 min) reduced uniformity of germination. The treatment had effect on morphology of seedlings that developed from the CP (7 min) and EMF (15 min) treated seeds resulting in reduced height/weight of stems and increased weight of leaves, respectively. Differential protein expression in the leaves of sunflower seedlings was assessed using 2D gel electrophoresis. Among the 104 differentially expressed proteoforms significantly higher abundance of 49 and 38 proteoforms and reduced abundance of 26 and 33 proteoforms was detected for the CP and EMF treated experimental groups, respectively. Hierarchical clustering revealed 3 major protein expression groups. Mass spectrometry analysis revealed several proteins involved in primary and protein metabolism or stress response, however chloroplast proteins with function involved in photosynthesis were dominant among the 40 identified proteoforms.
