Abstract
Šiame darbe buvo tiriama kaip skirtingomis sąlygomis vykstantis gliukozės metabolizmas veikia ląstelėse vykstančius energetinius procesus ir tetrafenilfosfonio (TPP+) jonų sugertį bei išmetimą iš L. lactis subsp. cremoris ląstelių. Nustatyta, kad TPP+ jonų sugerties ir išmetimo (ištekėjimo) iš ląstelių eiga priklauso nuo deguonies pasisavinimo intensyvumo, kuriam įtakos turi į terpę įneštos gliukozės kiekis bei hemino aktyvinamos kvėpavimo grandinės veikla. Tačiau hemino sukeliami pokyčiai ląstelėse nekeičia gliukozės sukeliamo TPP+ jonų judėjimo per bakterijų plazminę membraną pobūdţio. Be hemino augintos ląstelės pasiţymi intensyvesne glikolize. Terpės pH sumaţėjus iki 5 ir maţiau, slopinamas L. lactis ląstelių kvėpavimas, o silpstanti TPP+ jonų sugertis atspindi maţėjančią membranos įtampą. Su heminu augintos ląstelės atsparesnės rūgštiniam stresui. Intensyvėjant deguonies pasisavinimui, didėja TPP+ jonų sugertis ir ATP kiekis ląstelėse.
In this study it was investigated how different conditions of bacterial metabolism affect energetic processes and the flux of tetraphenylphosphonium cations (TPP+) through the cytoplasmic membranes of Lactococcus lactis subsp. cremoris cells. It was found that the flux of TPP+ through the cell membrane depends on oxygen absorbtion intensity, which in itself is affected by amount of glucose in medium and respiration chain activity induced by heme. However changes created in cells by heme does not affect the profile of TPP+ flux through cell membrane. Cells grown without heme have more intensive glycolysis, than those grown with heme. In mediums with pH 5 and lower, L. lactis respiration is inhibited and decreasing absorption of TPP+ ions indicates lowering membrane potential. Cells grown with heme have higher acidic stress resistance. When intensity of oxygen absorption gets higher, TPP+ ions absorption also get higher, as well as the amount of ATP in cells.
