Abstract
Kriosaugojimas arba biologinės medžiagos laikymas esant ypač žemai temperatūrai (-196ᵒC) yra vienas efektyviausių metodų ilgalaikiam augalų genetinių išteklių saugojimui. Šis metodas užtikrina saugų, ekonomišką ir ilgalaikį įvairių rūšių augalų genetinių išteklių išsaugojimą. Šiame tyrime buvo naudotos tabako (Nicotina tabacum L.) ir morkų (Daucus carota L.) veislės ‘Ieva’ ląstelių suspensijos auginamos LAMMC, Sodo augalų genetikos ir biotechnologijos skyriaus kolekcijoje in vitro. Tyrimas atliktas Lietuvos agrarinių ir miškų mokslų centro (LAMMC) filialo Sodininkystės ir daržininkystės instituto, sodo augalų genetikos ir biotechnologijos skyriuje, Kriobiologijos laboratorijoje, kuri taip pat priklauso Augalų genetikos ir biotechnologijos centrui. Šio tyrimo tikslas - nustatyti tinkamiausias kriosaugojimo sąlygas genetiškai modifikuotų morkų ir tabako ląstelių suspensijų ilgalaikiam saugojimui skystame azote (-196ºC temperatūroje).
Tiriamosioms morkų ir tabako ląstelių suspensijoms kriosaugoti buvo naudotos inkapsuliacijos - vitrifikacijos arba dehidratacijos metodikos, naudojant skirtingus krioprotektorius arba jų mišinius - sacharozę, glicerolį, PVS2 mišinį, sorbitolį bei DMSO. Tyrimų rezultatai rodo, kad genetiškai modifikuotoms tabako ląstelėms kriosaugoti tinka 80 min. ekspozicija 100 % PVS2 tirpale, o morkų ląstelėms - po 40 min. 50 % ir 100 % PVS2 tirpaluose. Naudojant krioprotektorius sacharozę ir glicerolį, tinkamiausia ekspozicijos trukmė tabako ląstelėms 1 valanda, morkų ląstelėms – 2 valandos. Parodyta, kad tabako ir morkų suspensijos ląstelėms kriosaugoti naudojant sacharozę ir glicerolį, tinkamiausias yra lėtas (2 val.) užšaldymas iki -40ºC, o naudojant PVS2 tirpalą - greitas užšaldymas SA. Nustatyta, kad didžiausias tabako ir morkų ląstelių gyvybingumas, saugant jas SA, siekiantis atitinkamai 63,7 ir 21,9 %, gautas naudojant inkapsuliacijos - vitrifikacijos metodą.

Cryopreservation or storage of biological material at extremely low temperatures (-196ᵒC) is one of the most effective methods for long-term conservation of plant genetic resources. This method ensures the safe, economical and long-term conservation of plant genetic resources of various species. Cell suspensions of tobacco (Nicotina tabacum L.) and carrot ((Daucus carota L.) cultivars ‘Ieva’) that were grown in the LAMMC, in vitro collection of the Department of Orchard Plant Genetics and Biotechnology, were used in this study. The study was performed at the Lithuanian Research Centre for Agriculture and Forestry (LAMMC), Department of Orchard Plant Genetics and Biotechnology, Laboratory of Cryobiology, which also belongs to the Center of Plant Genetics and Biotechnology. The aim of this study was to determine the most suitable cryopreservation conditions for long-term storage of genetically modified carrot and tobacco cell suspensions in LN (-196ºC).
Encapsulation - vitrification or dehydration techniques using different cryoprotectants or their mixtures - sucrose, glycerol, PVS2 mixture, sorbitol and DMSO were used to cryoprotect the carrot and tobacco cell suspensions. Studies show that 80 min. of exposure is suitable for cryopreservation of genetically modified tobacco cells in solution of 100% PVS2, and for carrot cells - 40 min. in solutions of 50 % and 100 % PVS2. When using cryoprotectants sucrose and glycerol, the most appropriate duration of exposure is 1 h for tobacco cells, 2 h for carrot cells. It has been shown that slow (2 h) freezing at -40ºC is most suitable for cryoprotection of tobacco and carrot suspension cells using sucrose and glycerol, and rapid freezing LN is suitable for using PVS2 solution. It is established that, the highest viability of tobacco and carrot cells when stored in LN, at 63,7 and 21,9 %, respectively, was found to be obtained by using the encapsuliation - vitrification method.
