Darbo tikslas - išgryninti lipopolisacharidus iš skirtingų gramneigiamųjų bakterijų, naudojant džiovinimo šalčiu ir karšto fenolio metodą, nustatyti jų koncentraciją spektrofotometriškai.
Šiame darbe lipopolisacharidų ekstrakcijai buvo naudoti džiovinimo šalčiu ir karšto fenolio metodai, o lipopolisacharidų koncentracija nustatyta spektrofotometrinės analizės būdu, kuris pagrįstas spalvine tiobarbitūrinės rūgšties reakcija su 2-keto-3-deoksioktano (KDO) rūgštimi.
Tyrimų metu buvo atlikta skirtingų gramneigiamųjų bakterijų lipopolisacharidų koncentracijos palyginamoji analizė ir lipopolisacharidų nustatymo spektrofotometriniu metodu optimizavimas. Taip pat įvertinta, kokią įtaką LPS išsiskyrimui daro skirtingos bakterijų kultivavimo sąlygos: bakterijų auginimas mažuose tūriuose ir fermentatoriuje, bakterijų auginimas 37℃ ir 40℃ temperatūroje.
Atlikus spektrofotometrinių tyrimų rezultatų analizę, galima teigti, jog didžiausia gryno lipopolisacharido masės dalis liofilizate buvo S. enterica ∆TolC ir S. enterica SL1344 bakterijose. KDO rūgšties nustatymo metodas buvo optimaliausias, kai buvo naudojamos tokios reagentų koncentracijos: 1,92 mM sieros rūgšties, 0,39 mM perjodato rūgšties, 9,75 mM natrio arsenato heptahidrato ir 12,3 mM tiobarbitūrinės rūgšties, o optimalus bakterijų skaičius suspensijoje buvo tuomet, kada KFV viename bakterijų suspensijos mililitre buvo lygus 1,29 · 1011. Auginant S. enterica SL1344 fermentatoriuje, LPS koncentracija suspensijoje ir supernatante buvo didesnė nei tada, kai jos buvo kultivuojamos mažuose tūriuose. Taip pat S. enterica SL1344 bakterijų LPS koncentracija buvo didesnė tada, kai bakterijos buvo auginamos 40℃, nei 37℃ temperatūroje.

Work aim - purify lipopolysaccharides from different gram-negative bacteria by freeze-drying and hot phenol methods, determine the concentration of lipopolysaccharides spectrophotometrically.
Freeze-drying and hot phenol methods were used for lipopolysaccharides extraction while concentration of lipopolysaccharides was determined using spectrophotometric analysis, based on the colour reaction between thiobarbituric acid with 2-keto-3-deoxy-D-mannooctanoic (KDO) acid.
The research consisted of comparative analysis between lipopolysaccharide concentration in different gram-negative bacteria as well as optimising spectrophotometric determination methods of lipopolysaccharides. Also, the effects of various cultivation conditions for the LPS concentration were evaluated – growing bacteria in small volumes and in the bioreactor, growing bacteria in 37℃ and 40℃ temperatures.
After conducting the analysis of results gathered from spectrophotometric research, we can claim that the largest LPS pure mass fraction in the lyophilizate was found in S. enterica ∆TolC and S. enterica SL1344 bacteria. KDO acid determination method proved to be the most optimal when the following reagents’ concentrations were used: 1.92 mM of sulfuric acid, 0.39 mM of periodate acid, 9.75 mM of sodium arsenate heptahydrate and 12.3 mM of thiobarbituric acid, while the optimal number of bacteria was when the CFU in one millilitre of bacterial suspension was 1,29 · 1011. LPS concentrations in the suspension and supernatant were larger when S. enterica SL1344 was grown in the bioreactor rather than when it was cultivated in small volumes. Also, when the S. enterica SL1344 bacteria were grown in 40℃ temperature, their LPS concentration was larger than in 37℃ temperature.
