Abstract
Disertacinio darbo tikslas – nustatyti hipertermijos (HT) ir hiperterminės chemoterapijos poveikio taikinius sveikų audinių mitochondrijose ir vėžinėse bei auglio neformuojančiose ląstelėse. Pirmą kartą įvertinti HT sukelti sveikų žiurkės smegenų ir kepenų izoliuotų mitochondrijų funkcijų bei jų fermentų aktyvumo pokyčiai bei atsako į HT stresą priklausomybė nuo lyties, kepenų mitochondrijose - ir nuo amžiaus. Gauti rezultatai, kad atsakas į HT priklauso nuo audinio, iš kurio buvo izoliuotos mitochondrijos, gyvūno lyties ir amžiaus. Pirmą kartą nustatyta, kad vienas iš HT poveikio taikinių sveikų smegenų ir kepenų audinių mitochondrijose yra glutamato dehidrogenazė (GDH), kurią HT ženkliai slopino. Palyginus GDH atsaką į HT įvairiose auglio neformuojančiose ir vėžinėse/auglį formuojančiose ląstelių linijose, nustatyta, kad auglio neformuojančių ląstelių GDH jautrus HT sąlygoms (kaip ir mitochondrijų iš sveikų audinių), tačiau vėžinėse/auglį formuojančiose ląstelių linijose HT sukėlė labai skirtingus GDH aktyvumo pokyčius – slopino, neturėjo poveikio arba net stimuliavo. Nustatyta stipri koreliacija tarp ląstelių GDH aktyvumo pokyčių HT streso metu ir ląstelių išgyvenamumo po HT. Gauti rezultatai, kad HT sustiprina citotoksinį cisplatinos poveikį kiaušidžių vėžio ląstelėms, ir šis poveikis susijęs su GDH fermento aktyvumo ir GDH raiškos pokyčiais.
The aim of the study was to determine the potential targets of hyperthermia (HT) and hyperthermic chemotherapy in mitochondria isolated from healthy tissues and in cancerous and non-tumorigenic cells. For the first time mitochondrial functions, enzyme activities and changes induced by HT were compared in mitochondria isolated from healthy rat brain and liver. Sexual dimorphism in the response to HT was evaluated in brain and liver mitochondria; the age dependency of this response was assessed in liver mitochondria. The obtained results revealed that mitochondrial response to HT depends on many factors: tissue, sex and age of the animal. It was shown for the first time that one of the HT targets in brain and liver mitochondria was glutamate dehydrogenase (GDH) enzyme, which was strongly inhibited by HT. For the first time the response of GDH to HT was compared in different non-tumorigenic and different types of cancer cell lines. The obtained results revealed that GDH in non-tumorigenic cells was inhibited by HT (like in mitochondria from healthy tissues), however, in the cancer cell lines HT induced changes in GDH activity was highly heterogeneous – inhibition, absence of effect or even stimulation. The correlation was established between the extent of GDH inhibition by HT and loss of cell viability. The novel fact was demonstrated that HT potentiates cytotoxic effect of cisplatin in ovarian cancer cells, and that the combinatory action of HT and cisplatin may be explained by the downregulated GDH expression.
